Enterococcusfaecalis is a nonmotile microbe, gram-positive, commensal bacterium inhabiting the gastrointestinal tracts of humans and other mammals,and ferments glucose without gas production, and does not produce a catalase reaction with hydrogen peroxide. It can produce a pseudocatalase reaction if grown on blood agar and a reduction of litmus milk, but does not liquefy gelatin. Also, It shows consistent growth throughout nutrient broth which is consistent with being a facultative anaerobe and catabolize a variety of energy sources including glycerol, lactate, malate, citrate, arginine, agmatine, and many keto acids. Enterococci survive very harsh environments including extremely alkaline pH (9.6) and salt concentrations.
The metabolism of the E. faecalis is mainly as:
1) Produce natural antibiotics, which is good for human/animals health, and bacteriostatic substances (bacteriocin) to inhibit the growth of pathogenic bacteria (E.coli and Salmonella) and improve the GI health.
2) Inhibit the reproduction of urease bacteria and putrefactive bacteria in the intestinal tract, reduce the content of urease and endotoxin in the intestinal tract, and reduce the content of ammonia and endotoxin in the blood.
3) Enhance the activity of macrophages, promote the immune response of animals, and increase the level of antibodies
4) Develop biofilms to adhere to the animal intestinal mucosa and reproduce, then forming a lactobacillus barrier to resist the side effects of pathogens and mycotoxins, which is bacillus, yeast and other pass- through bacteria could not work like this.
E. faecalis is also resistant to many commonly used antimicrobial agents (aminoglycosides, aztreonam, cephalosporins, clindamycin, the semisynthetic penicillins nafcillin and oxacillin, and trimethoprim-sulfamethoxa. So it can be used with some antibiotics to treat diseases. Also, it is a facultative anaerobic lactic acid bacteria and can resist bile salts, detergents, heavy metals, ethanol, azide, and desiccation and grow in the range of 10 to 45 °C and survive at temperatures of 60°C for 30 min, which is more suitable for production and application, compared with Bifidobacterial.

Enterococcus spp. has long been recognised for its probiotic benefits and is widely used around the world. One of the major benefits of Enterococcus spp. is that it is uniquely suited to survive the digestive process and flourish in the gut. It promotes a balanced gut environment by competing for
resources that harmful organisms would otherwise consume and use to grow. It also competes with harmful organisms for adhesion sites - areas on the surface of cells to which other cells and molecules can bind.
However, some strains of Enterococcus spp. maybe pathogenic and harmful to humans.
1)Virulence of Enterococcus spp. is not well understood, but adhesins, haemolysin, hyaluronidase, aggression substance and gelatinase are putative virulence factors. These virulence genes maybe transferred to human endogenous strains present in the gastrointestinal tract, which may contribute to increasing the virulence factors of this genus and endogenous strains.
2) Moreover, anti-microbial resistance (AMR) to many commonly used antibiotics has been reported, and E.faecium is the leading cause of multi-drug resistant enterococcal infections in humans. E. faecium resistance is acquired by gene transfer systems, such as conjugative and non-conjugative plasmids or transposons.
3) Although, there are conflicting views and different regulatory approaches to E. faecium around the world.health authorities all over the world are closely monitoring the development of AMR, including the use of E. faecium as a probiotic in food and feed.
In the USA, bacterial species that should not be used in dietary supplements include Escherichia coli, Enterococcusfaecalis and Enterococcusfaecium.Conversely, Enterococcus spp is accepted for use in animal feed by the Association of American Feed
Control Officials (AAFCO) as a direct fed microorganism, provided the microorganism is non-toxigenic. In China, CFDA does not approve this strain to be free used in human foods but some of the OTC drugs approved are with Enterococcusfaecium in China, this strain is mainly approved for animal health or animal feed application whatever by the direct-fed microbes or via their metabolism mechanism to benefits the animals or environments.
